Although the study of helminth-derived extracellular vesicles (EVs) is in its infancy, proteomic studies of EVs from representatives of nematodes, cestodes and trematodes have identified homologs of mammalian EV proteins including components of the endosomal sorting complexes required for transport and heat-shock proteins, suggesting conservation of pathways of EV biogenesis and cargo loading between helminths and their hosts. However, parasitic helminth biology is unique and this is likely reflected in helminth EV composition and biological activity. This opinion article highlights two exceptional studies that identified EVs released by Heligmosomoides polygyrus and Fasciola hepatica which display differential lipid and glycan composition, respectively, when compared with EVs derived from mammalian cells. Furthermore, we discuss the potential implications of helminth EV lipid and glycan composition upon helminth infection and host pathology. Future studies, focusing on the unique composition and functional properties of helminth EVs, may prove crucial to the understanding of host-parasite communication.Introduction To determine root canal cross-sectional shapes (RCCSS) of human permanent teeth using a new CBCT software. Methods RCCSS was determined on CBCT scans of 1,400 teeth (422 patients) as follows 1. circular; 2. conical-pyramidal; 3. oval/long oval; 4. flat/ribbon-like; 5. eight-shaped; 6. C-shaped; 7. calcified; 8. trapezoidal; 9. drop-shaped; 10. other shapes. Root canal shapes were evaluated in the coronal and middle thirds, 2 mm below their beginning, and in the apical third, 1 and 2 mm short of the apical foramen. Categorical variables were described as frequencies and percentages and analyzed using a chi-square test. The level of significance was set at p=0.05. Results Maxillary anterior teeth and maxillary first and second premolars had circular RCCSS at 1 mm and 2 mm from the apical foramen in more than 45% of the cases. The most frequent RCCSS in the buccal canal of maxillary first premolars at 1 mm from the apical foramen was circular (71%), followed by flat/ribbon-like (10%) and oval/long oval (6%). In mesiobuccal roots of maxillary and mandibular first molars at 1 mm from the apical foramen, the circular-shaped was found in 52% and 49%, respectively. Conclusions The root canal cross-sectional shapes in human permanent teeth are variable according to tooth group and root thirds. The highest frequency of circular-shaped canal at 1mm and 2mm from apical foramen was found in maxillary central incisors and mandibular first and second pre-molars. Oval-shaped canals were detected in practically all tooth group and root thirds.Xylem conduit diameter widens from leaf tip to stem base and how this widening affects the total hydraulic resistance (RTOT) and the gradient of water potential (Ψxyl) has never been thoroughly investigated. Data of conduit diameter of Acer pseudoplatanus,Fagus sylvatica and Picea abies were used to model the axial variation of RTOT and Ψxyl. The majority of RTOT (from 79 to 98%) was predicted to be confined within the leaf/needle. EHop016 This means that the xylem conduits of stem and roots, accounting for nearly the total length of the hydraulic path, theoretically provide a nearly negligible contribution to RTOT. Consequently, a steep gradient of water potentials was predicted to develop within the leaf/needle base, whereas lower in the stem water potentials approximate those of rootlets. In this review, we explore if TWEAK could provide a novel therapeutic strategy for managing respiratory disease in general and pulmonary arterial hypertension (PAH), obstructive sleep apnea syndrome (OSAS), asthma, idiopathic pulmonary fibrosis (IPF), chronic obstructive pulmonary disease (COPD) and non-small cell lung cancer (NSCLC), specifically.Background Besides SARS-CoV-2 RT-PCR testing, serological testing is emerging as additional option in COVID-19 diagnostics. Aim of this study was to evaluate novel immunoassays for detection of SARS-CoV-2 antibodies in human plasma. Methods Using EDITM Novel Coronavirus COVID-19 Enzyme Linked Immunosorbent Assays (ELISAs), we measured SARS-CoV-2 IgM and IgG antibodies in 64 SARS-CoV-2 RT-PCR confirmed COVID-19 patients with serial blood samples (n = 104) collected at different time points from symptom onset. Blood samples from 200 healthy blood donors and 256 intensive care unit (ICU) patients collected before the COVID-19 outbreak were also used. Results The positivity rates in the COVID-19 patients were 5.9% for IgM and 2.9% for IgG ≤ 5 days after symptom onset; Between day 5 and day 10 the positivity rates were 37.1% for IgM and 37.1% for IgG and rose to 76.4% for IgM and 82.4% for IgG after > 10-15 days. After 15-22 days the "true" positivity rates were 94.4% for IgM and 100% for IgG. The "false" positivity rates were 0.5% for IgM and 1.0% for IgG in the healthy blood donors, 1.6% for IgM and 1.2% for IgG in ICU patients. Conclusions This study shows high "true" vs. low "false" positivity rates for the EDITM SARS-CoV-2 IgM and IgG ELISAs.As cannabinoid use among the adolescent population becomes widespread with recent legalizations, understanding more about its effects on the developing brain becomes increasingly important. Adolescent cannabinoid use has been shown to elicit both short and long lasting effects on cortical function, in part due to its impact on maturing brain regions including the prefrontal cortex and associated inputs. This paper provides an overview of current state of knowledge on the lasting impact of repeated cannabinoid exposure on behavior and associated neural circuits in adolescents compared to other age groups. Data obtained from human and rodent literature are integrated to discuss potential neural mechanisms underpinning the enduring negative impact of cannabinoid exposure during this sensitive period of brain development.
