Here we review the clinical trials that specifically enrolled patients with BRAF-mutated CRC, from the phase I/II studies to the phase III trial BEACON CRC. We also examine the future directions towards a molecularly guided therapy for patients with BRAF-mutated CRC and the crucial role of a molecularly and clinically based algorithm in order to offer the best choice of treatment for these patients.Virtual clinical trials (VCT) are in-silico reproductions of medical examinations, which adopt digital models of patients and simulated devices. They are intended to produce clinically equivalent outcome data avoiding long execution times, ethical issues related to radiation induced risks and huge costs related to real clinical trials with a patient population. In this work, we present a platform for VCT in 2D and 3D X-ray breast imaging. The VCT platform uses Monte Carlo simulations based on the Geant4 toolkit and patient breast models derived from a cohort of high resolution dedicated breast CT (BCT) volume data sets. Projection images of the breast and three-dimensional glandular dose maps are generated for a given breast model, by simulating both 2D full-field digital mammography (DM) and 3D BCT examinations. Uncompressed voxelized breast models were derived from segmented patient images. Compressed versions of the digital breast phantoms for DM were generated using a previously published digital compression algorithm. The Monte Carlo simulation framework has the capability of generating and tracking ~105 photons/s using a server equipped with 16-cores and 3.0 GHz clock speed. The VCT platform will provide a framework for scanner design optimization, comparison between different scanner designs and between different modalities or protocols on computational breast models, without the need for scanning actual patients as in conventional clinical trials.In this work, we investigated microbiologically influenced corrosion (MIC) of X80 pipeline steel caused by nitrate-reducing bacteria Brevibacterium frigoritolerans (B. frigoritolerans) in an artificial Beijing soil using electrochemical measurements and surface analyses under aerobic conditions. The B. frigoritolerans was isolated from the surrounding soil of the X80 pipeline steel specimen in Beijing using culturing and molecular biology techniques. Confocal laser scanning microscopy images showed that the largest pit depth after 14 days due to B. frigoritolerans was approximately 7.16 μm. Electrochemical tests showed that the B. frigoritolerans could change the stability of the corrosion products on the 7th day. Inhomogeneous biofilm and the conductivity of Fe2O3 accelerated the corrosion process. Findings are mixed as to whether restrained eaters consume more food after exposure to unhealthy food marketing, and little is known about whether food marketing may have more impact on those who exercise successful dietary restraint as compared with those who experience dietary lapses, such as binge eating. In the current study, participants were 38 young women, ages 18-22 years old. Both dietary restraint and binge eating were measured by the Eating Disorder Examination-Questionnaire. Participants viewed both unhealthy food commercials and non-food commercials two separate times in the laboratory, and ad libitum candy intake was subsequently measured. Results indicated that participants who endorsed binge eating ate significantly more candy than those who did not endorse binge eating after they viewed unhealthy food commercials F (1, 35) = 20.49, p less then .001, η2 = 0.37, but not after viewing non-food commercials. No significant differences in candy eaten emerged when comparing those who endorsed dietary restraint as compared to those who did not, regardless of commercial type. Findings demonstrate the importance of specific operational definitions of restrained eating to consider the differences between those who report binge eating, and those who do not. They also suggest that individuals who engage in binge eating may be particularly susceptible to overeating in response to unhealthy food marketing, marking a possible area for intervention.Background and objective Mitochondrial diseases are a frequent cause of inherited genetic disorders caused by mutations in both the mitochondrial and nuclear human genome. The new technique of high-throughput sequencing, which is used more and more frequently around the world, is most often focused on nuclear DNA. In some cases, such data after proper IT processing could also allow to determine alterations in mtDNA genome. In our work, we want to verify that off-target reads from targeted gene panels are sufficient data to determine pathogenic variants in the mitochondrial genome. Methods We analyzed 50 patients who underwent routine diagnostics with the Illumina's TruSight One Sequencing Panel. In the entire bioinformatic analysis process, we have used only free, user-friendly and generally available online tools that do not require specialized IT knowledge. Results Most of the data obtained were suitable for determining the presence of homoplasmic variants in mtDNA; 84% of the data met the minimum 20-fold coverage requirement as defined in the scientific literature for clinical data.
