This review will discuss the mechanisms of Ca2+ dysregulation in AD and associated pathology and discuss potential approaches or strategies to develop novel drugs for the treatment of AD by targeting Ca2+ dysregulation.Background Noncoding RNAs (ncRNAs), such as microRNAs (miRNAs), small interfering RNAs (siRNAs) and long noncoding RNAs (lncRNAs), play significant regulatory roles in plant development and secondary metabolism and are involved in plant response to biotic and abiotic stresses. They have been intensively studied in model systems and crops for approximately two decades and massive amount of information have been obtained. However, for medicinal plants, ncRNAs, particularly their regulatory roles in bioactive compound biosynthesis, are just emerging as a hot research field. Objective This review aims to summarize current knowledge on herbal ncRNAs and their regulatory roles in bioactive compound production. Results and conclusion So far, scientists have identified thousands of miRNA candidates from over 50 medicinal plant species and 11794 lncRNAs from Salvia miltiorrhiza, Panax ginseng, and Digitalis purpurea. Among them, more than 30 miRNAs and five lncRNAs have been predicted to regulate bioactive compound production. The regulation may achieve through various regulatory modules and pathways, such as the miR397-LAC module, the miR12112-PPO module, the miR156-SPL module, the miR828-MYB module, the miR858-MYB module, and other siRNA and lncRNA regulatory pathways. Further functional analysis of herbal ncRNAs will provide useful information for quality and quantity improvement of medicinal plants.Introduction Resistance to corticosteroid is an essential mechanism in uncontrolled asthma as the corticosteroid is the mainstay of therapy. There are recent reports that epigenetic factors play a crucial role in the regulation of steroid action. Overexpression of P glycoprotein (P-gp) and reduce expression of Histone deacetylase 2 (HDAC2) has been linked to regulating the steroid action in other diseases like Nephrotic Syndrome (NS). However, their role in uncontrolled asthma is still not clear and warrant further investigation. We evaluated the expression and activity of P-gp and HDAC2 in patients with controlled asthma (CA) and uncontrolled asthma (UA). Methods A total of 60 CA (mean age 51.72±17.02 years, male=38), and 38 of UA (mean age=53.55±11.90 years, male=17) were recruited. The level of control was defined according to ( Global Initiative for Asthma) GINA 2016 criteria. The mRNA expression of HDAC2 and P-gp was studied by quantitative real-time polymerase chain reaction(PCR)the functional activity of P-gp was evaluated by commercially available kit via flow cytometry, and HDAC2 enzymatic activity was measured by commercially available kit by enzyme-linked immunosorbent assay (ELISA). Results P-gp expression and the functionality, was significantly higher in UA group of patients as compared to CA group of patients (p less then 0.005), moreover HDAC2 expression was significantly reduced in UA patients as compared to CA patients, (p less then 0.005). The enzymatic activity of HDAC2 was also significantly reduced in UA patients as compared to CA patients(p less then 0.005). Conclusion P-gp overexpression and HDAC2 under expression play an essential role in uncontrolled asthma by impairing the response to corticosteroid.and K. warburgiana are the only species of genus Kuehneola that parasitize plants belonging to genus Rosa (Rosaceae). Systematic revision of the genus Phragmidium and related genera by molecular phylogenetic analyses using nuc rDNA internal transcribed spacer (5.8S-ITS2 = ITS2) and nuc rDNA 28S (28S) sequences indicated that K. japonica and K. warburgiana belong to a group of Phragmidium species that also occur on Rosa. Morphological and molecular phylogenetic analyses revealed that these Kuehneola species were transferred to Phragmidium and renamed as P. japonicum and P. warburgianum. Two new Phragmidium species were also discovered on Rosa from China that could be distinguished from other species in the genus based on aeciospore or urediniospore morphology and phylogenetic placement. The first species, P. jiangxiense, is characterized by a urediniospore surface structure with stout spines that are basally embedded in the wrinkled spore wall. The second species, P. Metabolism inhibitor leucoaecium, is characterized by an aeciospore surface structure with irregularly elongated verrucae. Taxonomic descriptions and illustrations are provided.Purpose To validate the revised criteria of the International Workshop on Ocular Sarcoidosis (IWOS) for the diagnosis of ocular sarcoidosis (OS). Methods A retrospective chart review study was performed on 323 patients including 51 patients with biopsy-proven sarcoidosis and 272 patients with other uveitis entities. Data on intraocular signs and systemic investigations were collected, and sensitivity, specificity, positive predictive value (PPV), negative predictive value (NPV), and the Youden index were calculated. Results All intraocular signs and most systemic investigations showed high diagnostic parameters. Sensitivity, specificity, PPV, and NPV of the revised IWOS criteria were 1.000, 0.930, 0.728, and 1.000, respectively. Presumed or probable OS showed lower sensitivity and higher specificity when compared with the best Youden index. Conclusion The revised IWOS criteria are useful in Japanese patients, but could possibly be improved by modifying the criterion of presumed or probable OS.One in 10 children aged 11 to 16 years' have a mental health disorder. Whilst general practitioners (GPs) are considered ideal candidates to intervene and provide support, little research has focused on how this is managed, and the challenges they face, in the U.K. The aim of this work was to explore GPs' views on the management of adolescents with mental health disorders, and identify challenges they may face in current care. Eight GPs working in London were interviewed, using a semi-structured topic guide and a pre-prepared structured vignette. GPs with more clinical experience felt greater confidence in offering an intuitive approach to care. Many emphasised the therapeutic value of partnership with adolescents and discussed the complexity of identifying and labelling adolescents with a mental health disorder. Most GPs highlighted the need for a social picture assessing their level of socio-economic deprivation, and the impact of parents. However, through this focus, some GPs felt adolescents from middle class backgrounds could be overlooked.
