Lapatinib In a series of bioassays, insect mortality after specific exposure intervals to phosphine in selected species was examined, as well as the appearance of the so called "sweet spot". The species that were tested were Oryzaephilus surinamensis (L.), Tribolium castaneum (Herbst), Sitophilus oryzae (L.) and Rhyzopertha dominica (F.) with populations that had different levels of phosphine resistance. Evaluation was conducted by using the Phosphine Tolerance Test (PTT), with exposure of the adult stage for 15, 30, 60, 90, 150 and 300 min at 3000 ppm. At the end of these intervals (separate bioassays for each time interval), the insects were transferred to Petri dishes, in which recovery was recorded at different time intervals (2 h, 1, 2 and 7 days). The majority of susceptible populations of all species were instantly immobilized even in the shortest exposure period (15 min), in contrast with resistant populations that were active even after 300 min. After exposure to phosphine, populations and exposure time affected mortality of susceptible populations, whereas resistant populations recovered regardless of species and exposure time. Additional bioassays at the concentrations of 500, 1000, 2000 and 3000 ppm for 1, 3, 5, 20, 30 and 40 h showed the presence of the "sweet spot", i.e., decrease of mortality with the increase of concentration. In fact, for most of the tested species, the "sweet spot" appeared in 1000 and 2000 ppm at a 5-h exposure time, regardless of the level of resistance to phosphine. This observation is particularly important both in terms of the assessment of resistance and in the context of non-linear recovery at elevated concentrations, indicating the occurrence of strong hormetic reversals in phosphine efficacy.Erythropoietin (EPO) is a crucial hormone for erythropoiesis and produced by adult kidneys. Insufficient EPO production in chronic kidney disease (CKD) can cause renal anemia. Although hypoxia-inducible factors (HIFs) are known as a main regulator, the mechanisms of EPO production have not been fully elucidated. In this study, we aimed to examine the roles of retinoic acid (RA) in EPO production using EPO-producing cells derived from human induced pluripotent stem cells (hiPSC-EPO cells) that we previously established. RA augmented EPO production by hiPSC-EPO cells under hypoxia or by treatment with prolyl hydroxylase domain-containing protein (PHD) inhibitors that upregulate HIF signals. Combination treatment with RA and a PHD inhibitor improved renal anemia in vitamin A-depleted CKD model mice. Our findings using hiPSC-EPO cells and CKD model mice may contribute to clarifying the EPO production mechanism and developing efficient therapies for renal anemia.The orthoquartzite Imawarì Yeuta cave hosts exceptional silica speleothems and represents a unique model system to study the geomicrobiology associated to silica amorphization processes under aphotic and stable physical-chemical conditions. In this study, three consecutive evolution steps in the formation of a peculiar blackish coralloid silica speleothem were studied using a combination of morphological, mineralogical/elemental and microbiological analyses. Microbial communities were characterized using Illumina sequencing of 16S rRNA gene and clone library analysis of carbon monoxide dehydrogenase (coxL) and hydrogenase (hypD) genes involved in atmospheric trace gases utilization. The first stage of the silica amorphization process was dominated by members of a still undescribed microbial lineage belonging to the Ktedonobacterales order, probably involved in the pioneering colonization of quartzitic environments. Actinobacteria of the Pseudonocardiaceae and Acidothermaceae families dominated the intermediate amorphous silica speleothem and the final coralloid silica speleothem, respectively. The atmospheric trace gases oxidizers mostly corresponded to the main bacterial taxa present in each speleothem stage. These results provide novel understanding of the microbial community structure accompanying amorphization processes and of coxL and hypD gene expression possibly driving atmospheric trace gases metabolism in dark oligotrophic caves.Biomass gasification, especially distribution to power generation, is considered as a promising way to tackle global energy and environmental challenges. However, previous researches on integrated analysis of the greenhouse gases (GHG) abatement potentials associated with biomass electrification are sparse and few have taken the freshwater utilization into account within a coherent framework, though both energy and water scarcity are lying in the central concerns in China's environmental policy. This study employs a Life cycle assessment (LCA) model to analyse the actual performance combined with water footprint (WF) assessment methods. The inextricable trade-offs between three representative energy-producing technologies are explored based on three categories of non-food crops (maize, sorghum and hybrid pennisetum) cultivated in marginal arable land. WF results demonstrate that the Hybrid pennisetum system has the largest impact on the water resources whereas the other two technology options exhibit the characteristics of environmental sustainability. The large variances in contribution ratio between the four sub-processes in terms of total impacts are reflected by the LCA results. The Anaerobic Digestion process is found to be the main contributor whereas the Digestate management process is shown to be able to effectively mitigate the negative environmental impacts with an absolute share. Sensitivity analysis is implemented to detect the impacts of loss ratios variation, as silage mass and methane, on final results. The methane loss has the largest influence on the Hybrid pennisetum system, followed by the Maize system. Above all, the Sorghum system demonstrates the best performance amongst the considered assessment categories. Our study builds a pilot reference for further driving large-scale project of bioenergy production and conversion. The synergy of combined WF-LCA method allows us to conduct a comprehensive assessment and to provide insights into environmental and resource management.
